"Please join me in praying for the two National Guardsmen who were just shot moments ago in Washington D.C," Noem said via US social media company X.
She said the Department of Homeland Security is working with local law enforcement to gather more information.
The White House is "aware and actively monitoring this tragic situation," spokeswoman Karoline Leavitt said after the incident.
Leavitt noted that President Donald Trump, who is away for the Thanksgiving holiday in the state of Florida, was briefed on the shooting.
Separately, the DC Police Department said one suspect was in custody.
"MPD is on the scene of a shooting at 17th and I Street, NW. Please avoid the area," it said, adding the scene is secured.
The condition of the National Guard members who were shot has not yet been disclosed.
The Federal Aviation Administration reportedly said it had suspended departing flights to Ronald Reagan Airport, which serves the US capital, due to the security situation.
